WHAT IS SOFOSBUVIR?
Sofosbuvir is a nucleotide analog inhibitor of hepatitis C virus NS5B polymerase—the key enzyme mediating HCV RNA replication. The triphosphate form of sofosbuvir (GS-461203) mimics the natural cellular uridine nucleotide and is incorporated by the HCV RNA polymerase into the elongating RNA primer strand, resulting in viral chain termination.
Sofosbuvir is indicated for use by adults with chronic hepatitis C, meaning infection lasting more than six months. It is approved for people with HCV genotype 1, 2, 3, 4, 5 or 6. Sofosbuvir can be used by people being treated for hepatitis C for the first time (known as 'treatment-naive'). Sofosbuvir can be used by people with all stages of liver disease including compensated and decompensated cirrhosis. However, it works better for people with less advanced liver damage. People with chronic hepatitis C who are awaiting or have received a liver transplant tolerate sofosbuvir better than older medications.
POSSIBLE SIDE EFFECTS OF SOFOSBUVIR:
Get emergency medical help if you have any of these signs of an allergic reaction: hives; difficult breathing; swelling of your face, lips, tongue, or throat.
Call your doctor at once if you have:
pale skin, feeling light-headed or short of breath, rapid heart rate, trouble concentrating; or
fever, swollen gums, painful mouth sores, skin sores, cold or flu symptoms, cough.
Common side effects may include:
headache, tired feeling; mild itching; nausea; sleep problems (insomnia);
